Organic Amine Flash Purification Using a Novel Stationary Phase

A poster from Biotage presented at ACS 2015 in Boston shows how a unique amine-functionalized silica simplifies and improves purification of several basic tricyclic antidepressants with pKa values above 8 using a generic hexane/ethyl acetate gradient.

 

By neutralizing the acidic and protic silica surface with a bonded organic amine, basic compounds such as N-heterocycles, 2°, and 3° amines can be simply and efficiently purified with TLC-based elution data. The results from this research show that the chromatographic purification of organic amine compounds can be improved simply by using an amine-functionalized silica and typical normal-phase solvents such as hexane and ethyl acetate.
