
This application note describes the extraction of a panel of steroid hormones from human serum using ISOLUTE® SLE+ Supported Liquid Extraction plates prior to LC/MS-MS analysis. The simple sample preparation procedure delivers clean extracts and high, reproducible recoveries (>75%, RSD <10% ) for all analytes in human serum, with linearity >0.99 in the range 5–5000 pg/mL.

The method described in this application note achieves high recoveries of acrylamide in coffee. This application note describes the extraction and quantitation of Cannabimimetic Naphthoylindoles (Synthetic Cannabinoids) and their metabolites (JWH Series) from various matrices using Supported Liquid Extraction (SLE). Synthetic Cannabinoids or SPICE as they are commonly known have become an increasing problem as one of the newest forms of illicit drugs being consumed today. These compounds bind to the cannabinoid receptors in mammals triggering similar euphoric symptoms as Tetrahydrocannabinoids (THC). Currently robust and fast analytical methods of analysis are required to aid in the screening and detection of this growing class of compounds. The recoveries obtained for the synthetic cannabinoids parent and metabolites ranged from 70-98 %.

The use of schedule I drugs for patient pain management therapy warrants constant monitoring of therapeutic levels in the patient. Screening patient urine samples for the free drugs is complicated by the metabolism process which converts the free drug to the -glucuronide form. Patient urine samples can be enzymatically hydrolyzed and extracted to detect the drugs using Supported Liquid Extraction (ISOLUTE SLE+) which offers an efficient alternative to traditional liquid-liquid extraction (LLE) for bioanalytical sample preparation. This application note describes the extraction of testosterone and androstenedione from female patient serum samples using ISOLUTE® SLE+ (96 well) plates. This method has been optimized to extract testosterone and androstenedione from female clinical serum samples. The method has achieved very low limits of quantitation for testosterone and androstenedione 0.4nmol/L and 0.9nmol/L respectively. The recoveries for both analytes were greater than 90%.

Immunosuppressants inhibit or prevent the activity of the immune system used to prevent rejection of transplanted organs and treat autoimmune diseases. Due to the risks of immunodeficiency, those under treatment must undergo constant therapeutic drug monitoring (TDM) requiring reliable and robust analytical techniques for quantification of these drugs. Current methodologies use immunoassay techniques to measure immunosuppressant levels in patients which are expensive, time consuming and susceptible to issues with cross reactivity. The sample prep method in this application note offers an alternative approach to immunosuppressant analysis with LC-MS/MS giving more reliable and robust recoveries with no cross-reactivity and cost saving opportunities. Analyte recoveries range from 60-97% with RSDs all below 10%.

Vitamin D deficiency can result in a variety of health issues such as osteoporosis, liver and kidney problems, it is often associated with increased risk of cancers and multiple sclerosis. From this standpoint vitamin D monitoring is on the rise and of significant clinical relevance. This method describes the use of ISOLUTE SLE+ plates for the efficient and simple extraction of the vitamin D metabolites 25-OH vitamin D2 and 25-OH vitamin D3. Incorporated into the procedure is an integral protein binding disruption step which maximizes analyte recovery and eliminates the need for any offline protein disruption. This method has been internally validated using DEQAS approved serum samples and deuterated internal standards the results obtained are better than that required for criteria for acceptable performance for
DEQAS approval. All recoveries were consistently greater than 90% with all RSDs < 10% at LOQs of 1-3.75 ng/mL.

This application note details an optimized extraction protocol for the use of ISOLUTE SLE+ supported liquid 1 mL sample volume columns to extract a range of benzodiazepines from human urine. This simplified and efficient extraction method has analyte recoveries from 90% with LOQs as low as 40 ng/mL. The application has also been performed with halogenated and non-halogenated extraction solvents in the interests of providing for green chemistry.

Endogenous phospholipids present in biological fluids are a major problem in LCMS/ MS analysis as they are often very difficult to remove during sample preparation. When phospholipids are not removed, they retain very strongly on reversed phase analytical columns. If high organic (end of run) washes are not incorporated into the LC methods these matrix components may elute in subsequent analyses causing regions of suppression/enhancement leading to inaccurate quantitation. This poster evaluates the use of polymer-based solid phase extraction (SPE) sorbents, incorporating hydrophobic and various mixedmode retention mechanisms to address the problems associated with phospholipid removal.

Over the past decade, the need for non-invasive drug screening that that precludes sample adulteration has become attractive. As a result, detection using oral fluid devices for Drugs of Abuse (DOA) has come to the vanguard of the scientific community. The use of Supported Liquid Extraction (ISOLUTE® SLE+) prior to LC/MS or GC/MS can improve sample cleanliness without forfeiting sample detection within a diverse panel of DOAs. Here, we demonstrate the effects of altering elution solvent polarity and pH for sample pretreatment upon the simultaneous recovery of 34 compounds comprised of opioids, benzodiazepines, and stimulants to directly measure the effects of the oral fluid buffer, OraSure™, upon extraction and signal intensity at presumed LOQs.

Hair is a useful matrix for drug analysis, as it can be used to show historical drug use. Parent drugs are often present in high concentrations, and the matrix is stable over time.
This poster describes the use of the Biotage® Lysera to pulverise and extract hair samples, with ISOLUTE® SLE+ supported liquid extraction columns to clean up the samples prior to analysis. Limits of quantitation set by the Society of Hair Testing for amphetamines are achievable with the described method (0.2 ng/mg).

This poster demonstrates a rapid and reliable sample preparation method using Supported Liquid Extraction (ISOLUTE SLE+) to extract a suite of 30 hormone analytes from a hydrolyzed urine matrix. The ability to extract a broad range of different drugs from a biological matrix allows for the expedited analysis of a patient sample using LC-MS/MS. Typically small molecules are extracted from matrices like urine based on their polarities. A fast and reliable sample preparation method that could be implemented to extract drugs of different polarities from urine could be used as a screening tool to quickly identify the presence of illicit drugs in patient samples using LC-MS-MS.

This poster summarizes various sample preparation strategies for the
extraction of MMA from serum without the necessity for derivatization, prior to LC-MS/MS analysis. A range of sample preparation techniques of varying complexity were evaluated: protein precipitation, phospholipid depletion, supported liquid extraction and solid phase extraction using both silica and polymer-based mixed-mode anion exchange chemistries.
Method performance was evaluated for evaporative effects, assay recovery, ion suppression and phospholipid removal.

Immunosuppressant drugs are instrumental in preventing organ and tissue rejection in patients undergoing transplant surgery. Mycophenolic Acid is a common immunosuppressant drug used in patient transplant therapy. The ability to monitor the trough levels in patients to evaluate dosing is important for the administering of the drug. The free drug and its glucuronidated metabolite can be found in patient serum. The ability to extract the free drug and metabolite in patient serum is supported by using a fast and efficient sample preparation extraction method called Supported Liquid Extraction (ISOLUTE SLE+). The free drug and metabolite can be recovered from serum using ISOLUTE SLE+ with high enough efficiency to allow for quantitation at target trough levels.

Drug of abuse testing is typically conducted using urine specimens from patients being screened for illicit drug use. While collection of urine is considered a relatively non-invasive drug testing action, it is not convenient for law enforcement to do in the field. The ability to collect an oral fluid sample in the field can be considered non-evasive and simple to implement. Currently there are oral fluid collection kits (e.g. Immunalysis Quantisal™, Orasure Technologies Intercept®) that facilitate the collection in easy to follow protocols. Oral fluid samples collected using standard collection kits can be qualitatively and
quantitatively analyzed by LC-MS/MS, post extraction of the target analytes from the matrix. Drug analytes can be successfully extracted from oral fluid using Supported Liquid Extraction (ISOLUTE SLE+), which offers an efficient alternative to traditional liquid-liquid extraction (LLE). In this poster, we
demonstrate a new rapid and reliable sample preparation method to extract a broad suite of drugs (Figure 1) from neat oral fluid and buffered oral fluid matrices.

Antiepileptic drugs are prescribed to suppress seizures in epilepsy patients. A variety of different types of AEDs have been synthesized to pharmacologically address different types of epilepsy. The ability to therapeutically monitor these drugs in patients is necessary for maintaining optimal medical care and managing any adverse effects of thedrug. A fast and clean extraction method is needed that works in a variety of biological

Testing for drugs of abuse in oral fluids can strongly benefit the criminal justice field as a less invasive and cost-effective approach for drug detection when compared to blood or urine sampling. The Laser Diode Thermal Desorption (LDTD) source combined with Mass spectrometry is presented as a new screening tool for drug analysis in Oral Fluids. Analysis speed of LDTD provides accurate results in seconds in combination with exceptional specificity of MS instruments make a powerful platform for the screening of different drugs of abuse and new emerging drugs. Different extraction procedures are available; however those methods depend on specific drug conditions: basic or acid drugs / hydrophilic or hydrophobic. A new extraction approach, Supported Liquid Extraction (SLE+) is evaluated as generic extraction procedure.

Free urinary cortisol levels can be an indicator of a variety of disorders, such as Cushing Syndrome. This poster presents a simple method for the extraction of cortisol from urine demonstrating good recoveries and low ion suppression. LC-MS/MS analysis was performed using a Waters 2795 Liquid Handling Unit coupled to a Quattro Ultima Pt triple quadrupole MS using electrospray ionization, operated in the MRM mode. Supported liquid extraction method development resulted in recoveries greater than 90% for cortisol spiked urine. Calibration curves constructed using this method from 25-2000 ng/mL, demonstrated excellent linearity with coefficients of determination greater than 0.99.

Immunosuppressant drugs are extremely important for therapeutic drug monitoring (TDM) regimes, requiring robust and reliable methods for their analysis. This poster aims to demonstrate strategies for the extraction of various immunosuppressant drugs from whole blood providing clean samples prior to UPLC-MS/MS analysis. Spiked whole blood was extracted using a variety of protocols to investigate optimal combination of drug recovery and extract cleanliness. UPLC-MS/MS was conducted using a Waters ACQUITY UPLC coupled to a Quattro Premier XE triple quadrupole mass spectrometer. Acceptable extraction recoveries were obtained showing excellent extract cleanliness, demonstrating a reliable method for mass spectrometric approaches for these analytes.

Pain management therapy warrants constant monitoring of therapeutic levels of prescribed drug levels in patient urine samples. The number of samples being submitted for analysis has increased dramatically in the last 10 years with improvements in high throughput automated screening capabilities. Patient samples analysis is complicated by the need for an effective sample preparation methodology that can extract target analytes from complex matrices with good efficiency. Further complicating the process is the need to enzymatically hydrolyse the glucoronidated metabolites prior to extraction from the urine matrix. A fully automated sample preparation process using a TECAN Freedom EVO® 100 was designed to incorporate both the enzymatic hydrolysis and subsequent sample preparation assay as one continuous workflow. Supported Liquid Extraction (ISOLUTE SLE+) which offers an efficient alternative to traditional liquid-liquid extraction (LLE) and solid phase extraction (SPE) techniques was used to extract a suite of pain management drugs from spiked urine samples. A recovery and quantitation assay was run on the TECAN Freedom EVO® 100 using mock patient samples to demonstrate utility of automation process.

The use of schedule I drugs for patient pain management therapy warrants constant monitoring of therapeutic levels in patient urine samples. Screening patient urine samples is further complicated by the metabolic process which converts the free drug to the - glucuronide form. The target drugs in patient urine samples can be enzymatically hydrolysed and extracted using Supported Liquid Extraction (ISOLUTE SLE+) which offers an efficient alternative to traditional liquid-liquid extraction (LLE) and solid phase extraction (SPE) techniques typically used for this type of clinical sample preparation.

Endogenous phospholipids (outline structure shown in
Figure 1.) present in biological fluids are a major problem
in LC-MS/MS analysis. Due to their strong retention
characteristics in reversed phase chromatography
phospholipids tend not to elute as discrete peaks and are
often very difficult to separate from analytes of interest.
This co-elution often leads to areas of suppression or enhancement in the chromatogram which in turn can cause
quantitation issues. Supported liquid extraction (SLE) is an
analogous technique to traditional liquidliquid

How can I maximise the volume of raw sample that can be loaded onto an ISOLUTE SLE+ product?
Answer:
The standard sample pre-treatment recommendation for ISOLUTE SLE+ samples is a 1:1 (v/v) dilution prior to loading. However, to load a larger volume of raw sample without exceeding capacity, we recommend the addition of a smaller volume (e.g. 1:3 or 1:9 (v/v)) of a more concentrated buffer or pH control reagent.
For NEUTRAL analytes, such as steroids, sample dilution may not be required, as neutral analytes often extract well at physiological pH, so raw sample can often be loaded directly (with internal standard, as required).

How do I remove non-polar lipids and improve ISOLUTE SLE+ extract cleanliness for low level analysis?
Answer:
Once the sample is loaded and absorbed onto the ISOLUTE SLE+ column or plate, use a 2 step elution process. Selectively elute non-polar lipids with hexane (discard this fraction), then elute analytes of interest with a suitable solvent. In general, methods that are suitable for a ISOLUTE SLE+ plate with capacity for 400 µL sample (p/n 820-0400-P01) are transferable to the column format with equivalent capacity (in this case 820-0055-B (pack of 50 columns) or 820-0055-B-500 (bulk pack of 500 columns) using the same extraction protocol.
