
This application note describes a procedure for sample pre-treatment and extraction of THC and metabolites from human hair, using Biotage® Lysera for matrix pulverization of the sample prior to clean up using ISOLUTE® SLE+ supported liquid extraction.
Manual processing protocols were developed using the Biotage® PRESSURE+ 96 (plate format) or 48 (column format) positive pressure manifolds. For automated processing, protocols were developed using Biotage® ExtraheraTM.

This application note describes the fully-automated extraction of opiates from acid-hydrolyzed urine and non-hydrolyzed urine prior to GC/MS analysis. The methods were automated using Biotage® Extrahera™ configured for use with ISOLUTE® SLE+ supported liquid extraction columns.
Using the Biotage® Extrahera™, 24 hydrolyzed samples were extracted in under 31 minutes and 24 non-hydrolyzed samples were extracted in under 36 minutes. The limits of quantitation meet or exceed the sensitivity requirements set by SAMHSA and EWDTS for workplace testing applications.

This poster demonstrates that a large urine panel, comprised of 43 DOAs, from multiple drug classes, can be simultaneously screened by mixed-mode cation exchange SPE (using EVOLUTE EXPRESS CX 96 well plates) despite their disparate intermolecular traits, by thoughtfully selecting appropriate organic wash and elution conditions that simultaneously enable sample isolation and detection along with minimizing sample matrix effects.

Most drugs are excreted in urine as glucuronide conjugates. Hydrolysis using a beta-glucuronidase enzyme to convert the metabolites to their “free” form for analysis increases sensitivity. Red abalone (Kura Biotech), abalone (Campbell Scientific), and recombinant (IMCSzyme) beta-glucuronidase enzymes were evaluated to determine which provided the most complete hydrolysis of glucuronide metabolites without effecting the overall recovery of non-conjugated compounds.
EVOLUTE EXPRESS CX 96-well plates were used to extract hydrolysed urine samples, and the impact of th enzymes was compared.

Answer:
The majority of applications will transfer onto Extrahera. Some pre-treatment steps may not be automatable, and there may be limited instances where volumes are not compatible. It is advisable to gain a copy of a customer’s application/methods and discuss the requirement with a Biotage representative.

Extrahera has been designed mainly to process SLE and SPE but depending on what you want to achieve, you can program a method to only perform some steps, which then essentially turns the Extrahera into a liquid handler.

We need to avoid cross contamination at all times. How does Extrahera take care of that?
Answer:
Biotage has taken great care during the design of Extrahera to minimize the chances of cross contamination. The design incorporates a special flow-through waste plate under the extraction plate which prevents droplet splashing and contamination between samples from the Luer outlets of a plate or column. The design and robotic arm movement is programmed to minimize the chances of cross contamination at all times.
