Malachite green (MG) and crystal (gentian) violet (CV) are triphenylmethane dyes used in aquaculture as fungicides and ectoparasiticides. They are rapidly metabolised to the leucomalachite green (LMG) and leucocrystal (gentian) violet (LCV) metabolites that persist in fish tissue. MG and CV are banned in many countries due to their mutagenicity and carcinogenicity, but may still be used illegally. Monitoring methodologies must be able to detect the compounds at 1 ng/g (US) or 2 ng/g (EU). In order to meet these LODs for complex matrices such as oily fish, and the low UV absorbance of the leuco forms, LC-MS-MS is the analytical method of choice. This application note focuses on ease of use when compared to other methods, and describes a QuEChERS based approach incorporating the use of aluminium oxide in enhanced dispersive SPE to reduce the levels of fat in the final extract. Additional cleanup is provided by polymeric EVOLUTE SPE columns, and the resulting extract demonstrated low ion suppression. Analyte recoveries of 60-105%, with low rsds, are achieved. The method is also applicable to Brilliant green.

The use of schedule I drugs for patient pain management therapy warrants constant monitoring of therapeutic levels in the patient. Screening patient urine samples for the free drugs is complicated by the metabolism process which converts the free drug to the -glucuronide form. Patient urine samples can be enzymatically hydrolyzed to cleave glucuronide moiety and produce the free form of drug. The target analytes can them be extracted from the urine matrix using EVOLUTE CX solid phase extraction cartridges. LOQs range from 5 ng/mL to 0.5 ng/mL with recoveries above 80% and RSD <10%

This method demonstrates the extraction of Mephedrone (MCAT) from a range of biological fluids including plasma, urine and whole blood. This extraction is achieved through cation exchange using EVOLUTE CX, a mixed-mode resin-based SPE sorbent. Analytes were quantified using LC-MS/MS and recoveries were all above 90%.

This poster presents a method for detection of approximately 100 drugs and metabolites in urine using mixed-mode polymeric cation exchange solid phase extraction (SPE).
Four different glucuronidase enzymes and several incubation times and temperatures were evaluated to determine optimum hydrolysis conditions for seven commonly detected glucuronide metabolites. Samples were subjected to offline hydrolysis and SPE (EVOLUTE® EXPRESS CX, Biotage, Charlotte, NC) and results compared to an SPE plate that incorporates in-well hydrolysis (EVOLUTE HYDRO CX, Biotage).

Most drugs are excreted in urine as glucuronide conjugates. Hydrolysis using a beta-glucuronidase enzyme to convert the metabolites to their “free” form for analysis increases sensitivity. Red abalone (Kura Biotech), abalone (Campbell Scientific), and recombinant (IMCSzyme) beta-glucuronidase enzymes were evaluated to determine which provided the most complete hydrolysis of glucuronide metabolites without effecting the overall recovery of non-conjugated compounds.
EVOLUTE EXPRESS CX 96-well plates were used to extract hydrolysed urine samples, and the impact of th enzymes was compared.

Is there an expiry (‘best before’) date for ISOLUTE and EVOLUTE sample preparation products?
Answer:
There is no fixed expiry date for these products.
All ISOLUTE and EVOLUTE products are packaged in inner metal laminate barrier bags, which provide an effective barrier to oxygen and moisture penetration. Recommended storage conditions are standard (see below).

Browse more questions and answers >EVOLUTE® CX combines non-polar and strong cation exchange functionality for extraction of basic analytes from biological fluids and other aqueous samples through a dual, mixed-mode mechanism. This provides additional selectivity and clean-up for complex samples, with salts, phospholipids and proteins eliminated from extracts. Sorbent performance is not compromised by drying during processing. Available in a variety of formats, including modular and 96-well plates and SPE columns, and two different particle sizes (30 µm and 50 µm) depending on application requirements.
The sorbent consists of the EVOLUTE® backbone, surface modified with sulfonic acid groups.
EVOLUTE® CX sorbent is available in EVOLUTE® EXPRESS columns and 96-well plate formats for use with fast Load-Wash-Elute procedures. Eliminate traditional conditioning and equilibration and increase throughput without compromising performance.
EVOLUTE® sorbents have a narrow pore diameter, tailored to eliminate co-extraction of proteins from biofluids. Removal of proteins during sample preparation avoids transfer in to the analytical system, reducing matrix effects in LC-MS/MS, and avoiding contamination of the analytical column. This minimises the need for frequent backflushing and guard columns replacement.
Using EVOLUTE® CX, interferences including proteins, salts, non-ionisable molecules and phospholipids are removed during the interference elution step, ensuring extremely clean extracts (figure 2).

EVOLUTE® CX uses a generic procedure for extraction of basic analytes in aqueous matrices. The dual retention mechanism retains bases through a combination of non-polar and strong cation exchange interactions. The simple wash steps in the generic method remove >98% of phospholipids from plasma samples.
The combination of robust polymer chemistry, uniform coverage of sulfonic acid groups and optimum exchange capacity provide greater than 85 % recovery for a wide range of basic analytes.
EVOLUTE® CX should not be used for extraction of quaternary amines and other strong bases, or basic analytes that are unstable at basic pH (see EVOLUTE® WCX).

Narrow particle size distribution provides consistent flow characteristics for a variety of sample matrices. Optimized packing techniques eliminate channelling, which can lead to analyte breakthrough.
Even viscous biofluids can be loaded onto EVOLUTE EXPRESS CX products without the need for conditinoing or equilibration, using fast load-wash-elute SPE.
The surface characteristics of each batch of EVOLUTE® CX are quality controlled and tested to ensure consistency.
