Cyclosporine is an immunosuppressant used in organ transplants to overcome rejection. Following the transplant of a heart, liver, kidney or lung, a patient must be regularly tested for the rest of their lives. Blood testing intervals vary from 1 month to 6 months and this rapid reference method is ideal for batch tests with the advantage of no metabolite interfer-ence which is always found in immunological methods.

The removal of interferences and sample concentration are
essential steps in trace level detection of pesticides, sheep
dips, herbicides and PCBs in river water, wastewaters and
trade effluents. The following methods were developed by
the Environment Agency National Laboratory Service facility
at Nottingham using the Biotage RapidTrace for automated
solid phase extraction and the TurboVap II for evaporation.
The procedures are UKAS accredited and can reach detection
limits of 0.001 ug/Litre for some compounds.

The use of schedule I drugs for patient pain management therapy warrants constant monitoring of therapeutic levels in the patient. Screening patient urine samples for the free drugs is complicated by the metabolism process which converts the free drug to the -glucuronide form. Patient urine samples can be enzymatically hydrolyzed to cleave glucuronide moiety and produce the free form of drug. The target analytes can them be extracted from the urine matrix using EVOLUTE CX solid phase extraction cartridges. LOQs range from 5 ng/mL to 0.5 ng/mL with recoveries above 80% and RSD <10%

Melamine is traditionally used in making plastics, however, it’s low cost and high nitrogen content has led to exploitation in various sections of the food industry, most notably involving dairy products. Sustained melamine exposure can result in kidney stones and renal failure, with the young being most susceptible. This application note has been optimized on the RapidTrace+; an updated version of the current RapidTrace software which allows the system to process smaller bed heights than were previously possible. The application note shows recoveries of 88-90% with RSDs <10%.

Ethyl glucuronide (EtG, Figure 1.) is an ethanol metabolite formed by the glucuronidation of the parent molecule. The presence of EtG in urine and plasma is a useful and sensitive biomarker of alcohol intake in forensic toxicology. EtG can be detected at very low levels and is useful in distinguishing between ingested ethanol and that which is adsorbed through the skin from ethanol. This application note has been optimized on the RapidTrace+; an updated version of the current RapidTrace software which allows the system to process smaller bed heights than were previously possible. The application note shows analyte recoveries all above 97% with RSDs >10%.

Cyclosporine (Figure 1) is an immunosuppressant drug most significantly used in post-operative organ transplants to reduce the chance of rejection of donor organs by decreasing the activity of the patient’s immune system. Subsequent to an organ transplant, patients are tested at regular intervals for the presence of very low levels of the compound. Question:
My SPE technique is RapidTrace+. Is it possible to apply bigger sample volume (1000 mL) without prior evaporation?
Answer:
The maximum sample volume in standard operational mode is 40 mL limited to a 5.8 mL load each time. This is limited by the sample vial size and will only give you a 5 sample capacity.
Loading 40 mL of sample will be achieved by 6 x 5.8 mL then an additional 5.2 mL.
If you were to use one of the reagent lines as a source and use the condition function then it is possible to load more sample contained in an external vessel onto the SPE column via the reagent line. You will only be able to process a limited number of samples as dictated by other parameters within your SPE method as they will use reagent lines. Additionally you will not be able to load 1000 mL as you are limited by programming lines in a particular extraction procedure.
Essentially the RapidTrace is not really the right tool for the job. You would be better off considering the Biotage® Horizon 5000 depending on your application http://www.biotage.com/product-page/biotage-horizon-5000

Magnetically Encoded Racks enable the user to designate a specific method to a rack. When using magnetically encoded racks, the method does not have to be initiated via the controller software, because the module reads the magnets and automatically knows which method to run. The module runs the number of samples corresponding to the number of sample test tubes placed in the rack.
